Madekwe was born into an ethnically mixed family. Both her parents were both English. She was raised in a south London council estate, and went to The Brit School at age fourteen. She graduated with a BA in acting at the Royal Academy of Dramatic Art. While at the academy, she performed in numerous stage productions such as the King Henry V playing Princess Katherine and Wuthering Heights portraying Catherine Earnshaw.

Also, the ITV2 show Secret Diary of a Call Girl saw her in 2008 as Bambi. Madekwe was the main character on How to Lose Friends And Alienate Others. The comedy released in 2008. Some of her theatre credits include Little Sweet Thing (by Roy Williams) as well as Flight Path (by David Watson).
Madekwe's US debut came in 2009 with the thriller The Beautiful Life. The drama aired for just two episodes before being removed from The CW.
